چکیده

We examined further the ffects of angiotensin-11 and K+ on phospholipid metabolism in the adrenal zona glomerulosa. In adrenal zona glomerulosa cell preparations, physiological and supraphysiological concentrations of angiotensin-I1 (10"2-10-4 M) and K+ (0-16 m ~ ) elicited doseand time-dependent increases in aldosterone production and (32P)phosphate incorporation into chromatographically purified phosphatidic acid and phosphatidylinositol; the stimulatory effect on the phosphatidate-inositide cycle was relatively specific as there were no apparent changes in (32P)phosphate labeled in areas of the chromatogram containing the major phospholipids, phosphatidylcholine and phosphatidylethanolamine. Angiotensin-11 also stimulated the turnover of prelabeled (S2P)phosphatidylinositol in vitro, and treatment in vivo with a diuretic (provoking Na+ depletion and stimulation of endogenous angiotensin-11 secretion) was attended by 2-3-fold increases in phosphatic acid, phosphatidylinositol, and polyphosphoinositides, and lesser increases in other phospholipids. Thus, the stimulatory effect of angiotensin-I1 on (32P)phosphate labeling observed in vitro may be due to stimulation of two processes, uiz. phosphatidylinositol hydrolysis (with a subsequent increase in phosphatidate synthesis) and de novo synthesis of phosphatidic acid. Cycloheximide inhibited aldosterone production and blocked the angiotensin-11-induced increases in (32P)phosphate incorporation into phosphatidic acid and phosphatidylinositol. Thus, similar to our findings in studies of adrenocorticotropin hormone action in the zona fasciculata-reticularis, a labile protein appears to be required for the stimulation of phospholipid metabolism during angiotensin-I1 action in the zona glomerulosa. The present findings provide further evidence to support the possibility that phospholipids play an important role in the stimulation of aldosterone synthesis by angiotensin-11 and K+.

Effects of Potassium Loading and Other Stimulating Conditions on the Glomerular Zone of Fetal Adrenal Cortex of Rats

Effects of several conditions known to stimulate the zona glomerulosa of the adrenal cortex in the adult were examined on the fetal adrenal cortex of rats histochemically and electron microscopically. The stimulating conditions were the following; administration of Na restricted diet to the mother, administration of KCl or angiotensin II to the fetus, spironolactone or insulin to the mother. In...

Potassium and angiotensin II increase the concentrations of phosphatidic acid, phosphatidylinositol, and polyphosphoinositides in rat adrenal capsules in vitro.

We examined the effects of K+ and angiotensin II, the major regulators of aldosterone secretion, on phospholipid metabolism during incubation of glomerulosa-rich, adrenal capsules. Addition of increasing amounts of K+ and angiotensin II to the incubation media elicited progressive increases in corticosterone production and capsular concentrations of phosphatidic acid, phosphatidyl-inositol, and...

Control of adrenal cell proliferation by AT1 receptors in response to angiotensin II and low-sodium diet.

The effects of angiotensin II (ANG II), the angiotensin type 1 (AT1) receptor antagonist losartan, and low-sodium diet on rat adrenal cell proliferation were studied in vivo with immunocytochemistry. Both ANG II and low-sodium diet increased proliferation of endothelial cells of the zona glomerulosa. Losartan prevented ANG II-induced hyperplasia of glomerulosa cells but not the effects of a low...

Aspects of angiotensin action in the adrenal. Key roles for calcium and phosphatidyl inositol.

The steps between exposure of bovine adrenal glomerulosa cells to angiotensin and the stimulated increase in aldosterone production were studied in two ways. Binding of angiotensin to receptors, and hormone effects on phosphatidyl inositol turnover, 45Ca2+ fluxes, and aldosterone production were measured directly. Other potential intermediate steps were investigated indirectly by use of inhibit...

Distribution of extracellular signal-regulated protein kinases 1 and 2 in the rat adrenal and their activation by angiotensin II.

The adrenal gland of the rat is continuously regenerated through proliferation of a stem cell population in the outer part of the gland. To clarify the location of proliferative events within the adrenal gland, and the factors that stimulate them, rat adrenal capsule preparations, consisting of capsule, zona glomerulosa (ZG) and the outer zona fasciculata (ZF) were maintained in vitro under dif...
